Essence and tasks of the population-statistical approach
This method is widely used in medical genetics to study how normal and pathological genes are distributed in a specific territory.
A key stage of the work is thorough statistical processing of the obtained data. The main goal of researchers is to calculate the frequency of occurrence of various genes in the genotypes of the studied population. During the analysis, the proportions of the following are evaluated:
- Heterozygotes;
- Dominant homozygotes;
- Recessive homozygotes.
The theoretical basis for all calculations is the Hardy-Weinberg law. It is this law that makes it possible to correctly analyze allele frequencies of both normal and mutant genes in stable population groups.
Somatic cell genetics method
This approach is used for an in-depth study of heredity and variability, but using somatic cells rather than germ cells as the object of study. Research is conducted in vitro on tissue and organ cultures.
There are four main directions of work with cell cultures:
- Simple cultivation — maintaining cell viability outside the organism.
- Hybridization — artificial fusion of different cell types (e.g., creating a human-mouse hybrid). An important mechanism of this process is the gradual loss of chromosomes. By observing which traits disappear simultaneously with chromosomes, scientists establish linkage groups of genes.
- Cloning — obtaining genetically identical descendants from a single original cell. A classical example of application is the creation of a hybridoma. This is a cell hybrid resulting from the fusion of a normal lymphocyte and a tumor cell. A hybridoma is capable of infinitely producing monoclonal antibodies of strictly a single type.
- Selection — targeted selection of those cells that possess predefined properties required by the researcher.
Biochemical method
Biochemical analysis is based on the study of metabolism in the body. Specialists focus on enzymes, amino acids, and hormones. The main sphere of clinical application of the method is early diagnosis of hereditary pathologies, which is performed in the first months of a child's life.
All hereditary metabolic diseases are classified into three major groups:
- Carbohydrate metabolism disorders;
- Amino acid metabolism disorders;
- Lipid metabolism disorders.
Using this method, pathological conditions such as altered blood serum phenylalanine concentration, decreased (or complete absence of) thyroid function, as well as diabetes mellitus and a number of other disorders are successfully diagnosed.
Dermatoglyphics method
Dermatoglyphics deals with the study of papillary patterns located on human fingers, palms, and soles.
The main properties of these patterns are that they are highly individualized for each person and remain absolutely unchanged throughout their life. Due to these characteristics, the method finds application in two important areas:
- Determination of twin zygosity (helps distinguish monozygotic twins from dizygotic twins);
- Personal identification, which is especially in demand in forensic practice.
Briefly: answers to topical questions
What is the mathematical formula of the Hardy-Weinberg law for calculating genotype frequencies in a population?
The mathematical formula of the Hardy-Weinberg law for calculating genotype distribution in an ideal population is: $(p + q)^2 = p^2 + 2pq + q^2 = 1$. In this expression, the components of the formula reflect the following indicators:
- $p^2$ is the frequency of the homozygous dominant genotype ($AA$);
- $q^2$ is the frequency of the homozygous recessive genotype ($aa$);
- $2pq$ — frequency of the heterozygous genotype ($Aa$).
This distribution is based on the fact that the sum of the frequencies of the alleles themselves is always equal to one ($p + q = 1$).
What substances are used to induce cell fusion during hybridization?
To induce cell fusion during hybridization, the following are used:
- Sendai virus — used initially;
- Polyethylene glycol — a modern method that causes cell membrane fusion due to the presence of hydrophilic and hydrophobic groups in its composition.
These methods are described for cell hybridization in hybridoma technology.
What are the main types of papillary patterns distinguished during analysis in dermatoglyphics?
During analysis in dermatoglyphics, three main types of epidermal ridge papillary patterns are distinguished. These include:
- Loops — an element of the papillary pattern;
- Whorls — an element of the individual design;
- Arches — another type of pattern.
These patterns are most pronounced in thick skin (on the finger pads, palms, soles). Their nature is strictly individual and does not change throughout life, which allows the method to be used for personal identification and medical genetics.
Class and exam questions
What law is the population-statistical method based on?
Its theoretical basis is the Hardy-Weinberg law, which is used to analyze the frequency of normal and mutant alleles in a population.
What is a hybridoma and how is it obtained?
A hybridoma is the result of the fusion of a normal lymphocyte and a tumor cell (hybridization and cloning method). It is used to produce monoclonal antibodies of a single type.
What is the biochemical method used for in the clinic?
It is necessary for the early diagnosis of hereditary metabolic diseases (carbohydrate, amino acid, lipid) in the first months of the patient's life.
What properties of papillary patterns are used in dermatoglyphics?
Papillary patterns on the palms and soles are strictly individual and remain unchanged throughout life.
